Modification of human monocytes and macrophages by magnetic nanoparticles in vitro for cell-based delivery

The aim of the study was to develop a method for modification human monocytes/macrophages by iron oxide magnetic nanoparticles in vitro . Materials and methods Iron were obtained co-precipitation coated with thin SiO 2 layer polyethylene glycol 3000. Murine macrophage-like cell line RAW 264.7, primary monocytes macrophages incubated 1–24 hours. efficiency cellular uptake measured using ferrozine-based microcopy Perls’ Prussian blue staining. viability tested fluorescent flow cytometry SYTOX Green. Results. Incubation RAW264.7 cell, at concentration ˃ 5 µg/mL on rotator 1 hour 37 °С provides loading into > 99% cells. have no adverse effect viability. cells modified showed change migration activity. nanoparticle ˃50 pkg (Fe)/cell. Conclusion According proposed method, loaded proved viable, they retain ability migrate, therefore can be used as cell-based delivery systems tumor diagnostic therapy.
